MPK6 encodes a plant TEY-type mitogen-activated protein kinase that is predominantly cytoplasmic but dynamically associates with the plasma membrane and trans-Golgi network and, during mitosis, concentrates in puncta on the pre-prophase band and phragmoplast, consistent with roles in secretory trafficking and cell division plane control. The protein is a single, ~395 aa Ser/Thr kinase with the canonical MAPK fold (N- and C-lobes) and a conserved catalytic core that includes the activation-loop TEY motif required for phosphorylation-dependent activation; structural studies have resolved the kinase domain architecture and highlighted MAPK-specific regions (e.g., insert and L16 loop) that influence intrinsic activity. Functionally, MPK6 is a central node in MAPK signaling downstream of diverse biotic and abiotic cues, integrating pathogen and stress signals with developmental programs; it promotes hormone and defense outputs in part by directly phosphorylating 1-aminocyclopropane-1-carboxylic acid synthase isoforms (e.g., ACS2/ACS6) to stabilize these enzymes and elevate ethylene biosynthesis, and genetic analyses support additional roles in embryogenesis, root and reproductive development, and regulation of reactive oxygen species-linked responses.
