NADK (Nicotinamide Adenine Dinucleotide Kinase) is an enzyme crucial in the regulation of intracellular NADP+ levels, which are necessary for metabolic processes such as fatty acid and DNA synthesis. The human NADK gene is located on chromosome 1p36.33 and encodes a protein that primarily functions in the cytoplasm. NADK catalyzes the phosphorylation of NAD+ to NADP+, playing a significant role in maintaining the cellular redox state. Structurally, human NADK forms a dimeric organization, as revealed by its crystal structure, and includes four subunits (chains A-D) with specific colors indicating different chains. The enzyme's activity is regulated post-translationally, but its pathophysiological significance in humans remains underexplored. Multiple orthologs of the NADK gene are present in various organisms, illustrating its evolutionary conservation and importance across different species.
