Immunofluorescence analysis of Neuroligin-1 was performed using 70% confluent log phase SH-SY5Y cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. NLGN1 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of Neuroligin-1 was performed using 70% confluent log phase SH-SY5Y cells. Nuclei (Panel b: Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing Plasma membrane localization. Panel e represents MCF7 cells having no expression of Neuroligin 1. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

NLGN1 Antibody in Immunocytochemistry (ICC/IF) — NLGN1 Polyclonal Antibody detects Neuroligin 1 protein by immunofluorescent analysis. Sample: DIV14 rat E18 primary cortical neurons were fixed in 4% paraformaldehyde at RT for 15 min. Green: Neuroligin 1 protein stained by NLGN1 Polyclonal Antibody (Product # PA5-78648) diluted at 1:500. Red: beta Tubulin 3/ Tuj1, stained by beta Tubulin 3/ Tuj1 antibody [GT1338] diluted at 1:500. Blue: Fluoroshield with DAPI .

NLGN1 Antibody in Western Blot (WB) — Western blot was performed using Anti-NLGN1 Polyclonal Antibody (Product # PA5-78648) and a 110 kDa band corresponding to Neuroligin-1 was observed only in Mouse Brain, Rat Brain and Mouse Cerebellum which are reported to be positive and not in any other tissues like Mouse Mammary gland and Skeletal muscle. Tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Rat Brain (Lane 2), Mouse Cerebellum (Lane 3), Mouse Mammary gland (Lane 4) and Mouse Skeletal Muscle (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).An uncharacterized band of ~90 kDa was also observed in positive tissues.

NLGN1 Antibody in Western Blot (WB) — Western Blot using NLGN1 Polyclonal Antibody (Product # PA5-78648). Various tissue extracts (75 µg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with NLGN1 Polyclonal Antibody (Product # PA5-78648) diluted at 1:1,000.

NLGN1 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across tissues owing to their inherent genetic constitution. Relative expression of Neuroligin-1 was observed only in Mouse Brain, Rat Brain and Mouse Cerebellum which are reported to be positive and not in any other tissues like Mouse Mammary gland and Skeletal muscle using Anti-NLGN1 Polyclonal Antibody (Product # PA5-78648) in Western Blot.
