Antibody detects endogenous levels of fragment of activated Notch 1 resulting from cleavage adjacent to Val1744.
NOTCH1 (Cleaved Val1744) refers to the activated intracellular domain of the Notch1 receptor generated following proteolytic processing at valine 1744. NOTCH1 is encoded by the NOTCH1 gene and is synthesized as a single-pass transmembrane receptor that regulates cell fate decisions, differentiation, proliferation, and apoptosis. Upon ligand binding (such as Delta-like or Jagged family ligands), NOTCH1 undergoes sequential proteolytic cleavages, culminating in gamma-secretase-mediated cleavage within the transmembrane region. This cleavage at Val1744 releases the Notch1 intracellular domain (NICD), which translocates to the nucleus where it associates with transcriptional regulators such as RBPJ to activate target gene expression, including HES and HEY family genes. Detection of NOTCH1 cleaved at Val1744 is widely used as a marker of active Notch signaling in developmental biology, stem cell research, and cancer studies, where aberrant Notch pathway activation has been implicated in tumorigenesis and hematologic malignancies.
