Antibody detects endogenous levels of fragment of activated Notch2 resulting from cleavage adjacent to Asp1733.
NOTCH2 (Cleaved Asp1733) refers to the activated intracellular domain of the Notch2 receptor generated following gamma-secretase-mediated proteolytic cleavage at aspartic acid residue 1733. NOTCH2 is encoded by the NOTCH2 gene and functions as a single-pass transmembrane receptor in the conserved Notch signaling pathway, which regulates cell fate determination, differentiation, proliferation, and survival. Upon ligand engagement by Delta-like or Jagged family members on adjacent cells, NOTCH2 undergoes sequential proteolytic cleavages that culminate in intramembrane cleavage and release of the Notch2 intracellular domain (NICD2). The cleaved intracellular domain translocates to the nucleus, where it associates with the DNA-binding protein RBPJ and transcriptional co-activators to drive expression of target genes, including members of the HES and HEY families. Detection of NOTCH2 cleaved at Asp1733 is widely used as a marker of active Notch2 signaling in developmental biology, stem cell regulation, immunology, and cancer research.
