NRT1.1 encodes a plasma-membrane, proton-coupled nitrate transceptor of the plant NRT1/PTR (NPF) family in Arabidopsis thaliana, enriched in specific cell types and developmental zones including root tips and lateral root primordia as well as guard cells, consistent with roles in nutrient acquisition and signaling. The protein adopts the canonical major facilitator superfamily fold with 12 transmembrane helices organized into two six-helix bundles that form a central transport pathway; structural studies indicate it can form homodimers in the membrane, and a key regulatory phosphorylation at Thr101 (near the dimer interface) shifts the transporter between low- and high-affinity nitrate uptake modes by altering dimerization/structural flexibility. Functionally, NRT1.1 mediates nitrate influx across the plasma membrane over a wide external concentration range and also acts in nitrate sensing/signaling to shape transcriptional nitrate responses and root system architecture (notably lateral root proliferation in nitrate-rich patches); in leaves, its activity in guard cells contributes to nitrate accumulation and electrical changes associated with stomatal opening, influencing transpiration and drought responses.
