MA1-10029 detects Nsp1p from S. cerevisiae samples. To confirm a high affinity for Nsp1p, strain RS453, which expresses a shortened isoform of Nsp1p, was compared to the wildtype strain BJ5465. NSP1 in the RS453 strain contains an internal deletion that removes the coding sequence for 6 FXFG repeats, which are not essential for protein function, and encodes a protein that has been observed to migrate at ~85kDa on SDS-PAGE gels. Wildtype Nsp1p has a predicted MW of 86.5 kDa but has been observed to migrate on SDS gels at ~100 kDa. SDS-PAGE testing results showed wild type Nsp1p at ~108 kDa and Nsp1p RS453 at ~91 kDa, confirming specificity for the Nsp1p protein.
For western blots of yeast protein samples, use MA1-10029 diluted 1:10,000 (cell lysates) to 1:50,000 (nuclear fractions), followed by chemiluminescent detection (ECL). For other (non-ECL) western detection methods, try MA1-10029 diluted 1:1,000 to 1:5,000. For immunofluorescence on yeast cells, use MA1-10029 diluted 1:1,000 to 1:10,000.
Nucleoporin NSP1 is a component of the yeast nuclear pore complex (NPC). NPC components, collectively referred to as nucleoporins (NUPs), can play the role of both NPC structural components and of docking or interaction partners for transiently associated nuclear transport factors. Active directional transport is assured by both, a Phe-Gly (FG) repeat affinity gradient for these transport factors across the NPC and a transport cofactor concentration gradient across the nuclear envelope (GSP1 and GSP2 GTPases associated predominantly with GTP in the nucleus, with GDP in the cytoplasm). NSP1 plays an important role in several nuclear transport pathways including poly(A)+ RNA, tRNA, pre-ribosome, signal recognition particle (SRP), and protein transport.
