Natamycin is a polyene macrolide antibiotic with strong antifungal properties, produced by Streptomyces natalensis. It functions by binding to ergosterol in fungal cell membranes, disrupting membrane integrity and inhibiting fungal growth and reproduction. Natamycin is commonly used as an antifungal food preservative due to its ability to prevent mycotoxin formation. Structurally, it is characterized by a large macrolide ring and conjugated double bonds, which contribute to its antifungal activity. Gene engineering has enabled the creation of natamycin analogs with enhanced properties, by modifying post-polyketide synthase tailoring enzyme encoding genes. This antibiotic is also used in medical applications, such as treating fungal keratitis, where its formulation challenges are addressed by developing natamycin-loaded nanomicelles to improve corneal permeability and reduce irritation.
