NdhS encodes a small, nucleus-encoded accessory subunit of the chloroplast NADH dehydrogenase-like (NDH) complex that localizes to thylakoid membranes on the stromal side, where it contributes to the hydrophilic, stroma-exposed arm of NDH. Structurally, NdhS belongs to a conserved NdhS protein family found in oxygenic photosynthetic lineages and is thought to act primarily through protein-protein interactions rather than catalysis, helping organize the electron-donor interface of the complex; specifically, biochemical and genetic evidence supports a role for NdhS in forming or stabilizing a ferredoxin interaction site required for high-affinity ferredoxin binding. Functionally, by promoting efficient ferredoxin-dependent reduction of plastoquinone, NdhS supports NDH activity in cyclic electron transport around photosystem I and chlororespiration, processes that augment proton motive force across the thylakoid membrane and thereby help tune ATP production and protect photosystems under fluctuating or high-light conditions.
