NeuGc-GM3 is a sialic acid-containing ganglioside in which the GM3 glycosphingolipid carries N-glycolylneuraminic acid (Neu5Gc) instead of the more common N-acetylneuraminic acid (Neu5Ac). While most normal human tissues lack Neu5Gc due to an inactive CMAH enzyme, NeuGc-GM3 is frequently detected in certain tumors, including melanoma, breast cancer, and lung cancer. Because of its limited expression in healthy human tissues and elevated presence in malignancies, NeuGc-GM3 is considered a tumor-associated antigen and has been investigated as a target for cancer immunotherapy and antibody-based treatments.
