NtcA is a cytosolic, sequence-specific DNA-binding transcription factor found in cyanobacteria, where it functions as a global regulator coordinating the cellular response to nitrogen availability. It is a CRP-family regulator that forms homodimers; each subunit contains an N-terminal effector-binding domain and a C-terminal helix-turn-helix DNA-binding domain connected by a long alpha-helical linker that helps position the DNA-recognition helices appropriately for promoter binding. NtcA directly controls expression of numerous genes involved in nitrogen uptake and assimilation (including activation of key assimilation functions and autoregulation), and its activity is tuned by the nitrogen starvation signal 2-oxoglutarate, which acts as an allosteric effector that enhances NtcA-DNA binding and promotes transcriptional activation of NtcA-dependent promoters under nitrogen limitation, thereby coupling carbon/nitrogen balance to transcriptional output.
