PFDN1 encodes prefoldin subunit 1, a soluble cytosolic protein that assembles with five partner subunits into the canonical eukaryotic prefoldin heterohexamer, a jellyfish-like complex built from paired beta-barrel cores with six extended coiled-coil "tentacles" whose distal hydrophobic surfaces capture non-native polypeptides. As part of this complex, PFDN1 functions as a molecular co-chaperone that binds and stabilizes newly synthesized or partially unfolded client proteins-classically including actin and tubulin-and promotes their productive folding by delivering them to the cytosolic chaperonin CCT/TRiC, thereby supporting proteostasis and proper formation of the actin- and microtubule-based cytoskeleton.
