Western blot analysis of PDK4 in, Lane 1: human HepG2 whole cell lysates, Lane 2: human Hela whole cell lysates, Lane 3: human A549 whole cell lysates, Lane 4: rat skeletal muscle tissue lysates, Lane 5: rat heart tissue lysates. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. PDK4 (PDHK4), along with the other PDHKs, regulates glucose metabolism by phosphorylating the E1 alpha subunit of the mitochondrial pyruvate dehydrogenase complex. Inhibition of PDHKs is viewed as a potential therapeutic treatment for type II diabetes. This protein is located in the matrix of the mitrochondria and it's expression is regulated by glucocorticoids, retinoic acid and insulin. PDK4 inhibits the mitochondrial pyruvate dehydrogenase complex by phosphorylation of the E1 alpha subunit, thus contributing to the regulation of glucose metabolism.
