PDLP1 encodes a plasmodesmata-localized, type I membrane protein that is delivered to the plasma membrane at plasmodesmata via the secretory pathway and is oriented with an N-terminal apoplastic/extracellular region and a short C-terminal cytosolic tail separated by a single transmembrane helix. Its ectodomain contains a signal peptide and two tandem DUF26 (cysteine-rich) domains, consistent with receptor-like features, while correct plasmodesmal targeting depends strongly on the transmembrane region. Functionally, PDLP1 modulates intercellular (cell-to-cell) molecular trafficking through plasmodesmata, thereby influencing symplastic connectivity; it also contributes to plant defense, including regulation of callose-associated responses and immunity during pathogen challenge, where PDLP1 can relocalize to haustoria-associated extrahaustorial membranes and promotes resistance phenotypes when overexpressed.
