PGRL1A encodes a chloroplast thylakoid-membrane protein that localizes to the photosynthetic electron-transfer membranes, where it physically associates with photosystem I and forms a functional complex with PGR5 to support cyclic electron flow around photosystem I. Structurally, PGRL1A is an integral membrane protein with transmembrane segment(s) anchoring it in the thylakoid and a cysteine-rich region important for redox chemistry; biochemical work indicates that conserved redox-active cysteines and an iron-containing cofactor are required for its activity and regulation. Functionally, the PGR5-PGRL1A pathway mediates antimycin A-sensitive cyclic electron flow by channeling electrons from ferredoxin back to the plastoquinone pool (i.e., ferredoxin:plastoquinone reductase activity attributed to PGRL1), thereby enhancing proton motive force generation without net NADPH production, helping tune the ATP/NADPH balance and contributing to photoprotection under fluctuating or high light by supporting lumen acidification-dependent control of electron transport.
