PIN2 is a plant auxin efflux carrier that directs polar auxin transport in roots and shoots, shaping growth responses such as gravitropism, root elongation, and lateral root patterning. It localizes to the plasma membrane with asymmetric distribution that is dynamically regulated by endocytosis, recycling, and phosphorylation, enabling directional auxin flow. PIN2 activity integrates signals from light, gravity, and hormones including ethylene and cytokinin, and its trafficking depends on clathrin mediated endocytosis and ARF GEF regulated recycling. Mutations in PIN2 alter auxin gradients, causing defects in root bending, epidermal cell expansion, and overall root architecture. PIN2 works with other PINs and ABCB transporters to coordinate tissue level auxin distribution and adaptive growth under environmental stress.
