PIP2-1, PIP2-2, PIP2-3, PIP2-4, PIP2-5, PIP2-6, and PIP2-7 encode members of the plant plasma membrane intrinsic protein (PIP) aquaporin subfamily that predominantly localize to the plasma membrane, where they assemble as tetramers in which each monomer forms an independent channel. Structurally, PIP2 proteins are typical major intrinsic proteins with six transmembrane helices connected by five loops, two conserved NPA motifs in re-entrant loops that meet within the membrane, and an aromatic/arginine selectivity filter that helps define substrate permeability; they often carry regulatory phosphorylation sites in cytosolic terminal regions and can be gated by cellular conditions such as cytosolic acidification. Functionally, PIP2 isoforms act as high-capacity pathways for transmembrane water movement that support tissue hydraulics and rapid water fluxes during development and environmental responses, and specific family members can also facilitate diffusion of small neutral solutes such as hydrogen peroxide, contributing to redox signaling and stress physiology; in several systems, water permeability and gating behavior are further modulated by co-expression and hetero-oligomerization with related PIP aquaporins, enabling dynamic control of membrane transport capacity.
