Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

Protochlorophyllide oxidoreductase is a chloroplast enzyme that catalyzes the light-dependent reduction of protochlorophyllide to chlorophyllide, a pivotal step in chlorophyll biosynthesis during greening. POR belongs to the short chain dehydrogenase/reductase family and uses NADPH as a hydride donor; its catalytic cycle is uniquely photoreversible, requiring the formation of a ternary POR NADPH protochlorophyllide complex that is activated by light to drive the reduction. Many angiosperms encode multiple POR isoforms (e.g., PORA, PORB, PORC) with distinct expression profiles across development and light conditions, enabling fine control of chlorophyll production during de etiolation and in mature tissues. POR is abundant in etioplasts, where it associates with prolamellar bodies, and its activity governs the safe buildup and conversion of tetrapyrrole intermediates, minimizing photooxidative stress. Perturbations in POR expression or function lead to defects in chlorophyll accumulation, impaired thylakoid development, light sensitivity, and compromised photosynthetic capacity, highlighting its central role in establishing a functional photosynthetic apparatus.
