PPDK encodes pyruvate, phosphate dikinase, a predominantly cytosolic enzyme in many organisms (and, in photosynthetic plants, also found in plastids of mesophyll cells) that functions as a reversible metabolic "energy-coupling" catalyst at the pyruvate/phosphoenolpyruvate node. The protein is a large, multi-domain phosphotransferase built from three major regions: an N-terminal nucleotide-binding domain that carries out the ATP/AMP partial reaction, a central phosphoryl-carrier (His) domain containing a conserved, transiently phosphorylated histidine essential for catalysis, and a C-terminal PEP/pyruvate-binding domain with an alpha/beta-barrel-type active-site architecture for binding pyruvate/PEP with divalent metal support. These reaction centers are spatially separated, and catalysis proceeds via a swiveling movement of the central His domain that shuttles the phosphoryl group between the N- and C-terminal active sites. Biochemically, PPDK catalyzes the interconversion of ATP + inorganic phosphate + pyruvate with AMP + pyrophosphate + phosphoenolpyruvate, enabling ATP-dependent PEP formation for carbon fixation support in C4 photosynthesis and gluconeogenic/PEP-supplying roles in diverse bacteria and protists, while also operating in the reverse direction in some microbes to support ATP formation depending on metabolic context.
