PPH1 encodes a chloroplast-localized, thylakoid-associated serine/threonine protein phosphatase that resides predominantly in the stroma lamellae, positioning it to access antenna proteins that reversibly migrate between photosystems during acclimation to changing light. The protein is a monomeric PP2C-type phosphatase synthesized with an N-terminal chloroplast transit peptide and a conserved PP2C catalytic core that coordinates a binuclear divalent-metal center (Mn2+ or Mg2+) typical of PP2C enzymes; structural work further shows additional surface motifs that create substrate-recognition clefts enabling selective binding of phosphorylated N-terminal segments of LHCII components such as Lhcb1. Functionally, PPH1 is the principal phosphatase responsible for dephosphorylating major LHCII proteins (notably Lhcb1/Lhcb2) while showing limited activity toward photosystem II core phosphoproteins, thereby antagonizing the STN7 kinase pathway and promoting the transition from state 2 to state 1. Genetic loss of PPH1 leads to LHCII hyperphosphorylation, increased effective antenna association with photosystem I, and a pronounced defect in state transitions, underscoring its central role in balancing excitation energy distribution and maintaining photosynthetic redox homeostasis under fluctuating light.
