PPR (pentatricopeptide repeat) proteins are predominantly organelle-targeted, helical repeat RNA-binding factors that localize mainly to mitochondria (typically the matrix, with some members also reported in the nucleus) and act on post-transcriptional steps of gene expression. In mammals, a small set of nuclear-encoded PPR-motif proteins support mitochondrial transcript handling; the best-characterized example is LRPPRC, a leucine-rich protein containing multiple tandem PPR motifs that form an extended alpha-solenoid scaffold suited for sequence-selective RNA interactions and assembly of ribonucleoprotein complexes. Functionally, LRPPRC partners with the RNA-binding protein SLIRP to bind mitochondrial mRNAs cotranscriptionally, protect them from 3-prime exonucleolytic decay, promote appropriate 3-prime polyadenylation, and facilitate delivery and efficient translation of specific mitochondrial transcripts on the mitoribosome, thereby sustaining oxidative phosphorylation capacity (notably including effects on cytochrome c oxidase-related gene expression); disruption of this PPR-dependent mRNA stabilization/translation axis is a recognized mechanism underlying severe mitochondrial disease phenotypes.
