Phosphoribulokinase is a chloroplast enzyme in plants that regenerates ribulose 1,5 bisphosphate in the Calvin cycle by phosphorylating ribulose 5 phosphate using ATP. It is essential for sustaining carbon fixation and photosynthetic efficiency, and its activity is regulated by light via thioredoxin mediated redox control and by metabolite levels that reflect stromal energy status. Phosphoribulokinase forms part of the photosynthetic enzyme network coordinating flux through the Calvin cycle, and changes in its expression or activation influence growth, biomass accumulation, and tolerance to environmental stress. Mutations or reduced activity limit ribulose 1,5 bisphosphate supply, slow CO2 assimilation, and disrupt redox balance and carbohydrate partitioning.
