PSMA3 encodes a ubiquitously expressed, soluble proteasome alpha-type subunit that localizes predominantly to the cytosol and nucleus as a core constituent of the 20S proteasome (and, by extension, the 26S proteasome). The gene produces alternatively spliced transcripts that yield distinct protein isoforms, and the canonical product is a ~28 kDa, ~255 amino-acid member of the peptidase T1A family that adopts the conserved proteasome alpha-subunit fold and assembles into the outer heptameric alpha rings of the 20S core particle, where alpha-subunit N-termini contribute to formation of the gated entry pore controlling substrate access to the proteolytic chamber. Functionally, PSMA3 supports ubiquitin/ATP-dependent protein turnover by helping build and maintain the architecture of the 20S core that partners with regulatory particles to degrade polyubiquitinated substrates, thereby shaping proteostasis, cell-cycle progression, and stress responses; consistent with this role, PSMA3 has also been reported to bind diverse cellular proteins, linking the proteasome to broader regulatory pathways including aspects of mRNA metabolism.
