PSB33 encodes a nucleus-derived chloroplast protein that is imported via an N-terminal transit peptide and accumulates as an integral component of the thylakoid membrane, where it associates with photosystem II (PSII)-light-harvesting complex II (LHCII) assemblies. In Arabidopsis, PSB33 is a small membrane protein (approximately 23 kDa by immunodetection) predicted to contain a single transmembrane helix that harbors an LHC-binding motif, plus a comparatively large surface-exposed head domain positioned to interact with PSII/LHCII components; biochemical fractionation and complex analyses place it near LHCII and the PSII core antenna subunit CP43. Functionally, PSB33 acts as an auxiliary PSII core factor that supports the stability and/or organization of PSII-LHCII supercomplexes and helps optimize photosynthetic performance under changing or stressful light conditions: loss-of-function mutants show reduced PSII-LHCII supercomplex accumulation, strongly impaired state transitions, reduced nonphotochemical quenching capacity, and heightened photosensitivity (including under UV-A), consistent with a role in maintaining a functional PSII pool during light stress.
