Pygopus 1 (PYGO1) is a gene located on chromosome 6p21.31, encoding a protein that plays a crucial role in the Wnt signaling pathway, which is fundamental for various developmental processes including cell proliferation, polarity, and fate determination. PYGO1 is a part of the Pygopus family of proteins, characterized by their Plant Homeodomain (PHD) finger domain that facilitates interaction with histones. Within the Wnt signaling pathway, PYGO1 acts as a coactivator by binding to components of the transcription complex, thus aiding in the expression of Wnt target genes. The protein is especially important in embryonic development and is implicated in specific processes such as limb formation and neural development. Dysregulation of PYGO1 and the Wnt pathway is associated with developmental disorders and certain cancers, demonstrating the protein's significance in maintaining cellular homeostasis. Current studies are focusing on the mechanistic aspects of PYGO1's function in transcription regulation and its potential role in therapeutic strategies targeting Wnt signaling-related diseases.
