Paromomycin is an aminoglycoside antibiotic derived from Streptomyces rimosus, exhibiting broad-spectrum activity against various infections such as amoebiasis, cryptosporidiosis, giardiasis, and numerous gram-negative and gram-positive bacteria. Its structure is characterized by a complex arrangement of sugar moieties, which contribute to its binding affinity and mechanism of action, primarily targeting the bacterial ribosome to inhibit protein synthesis. Paromomycin's gene location and modifications have been explored to enhance its efficacy and overcome resistance, with targeted chemical syntheses revealing efficient modifications within the neomycin family. These modifications aim to convert neomycin and its derivatives into more potent forms, addressing the growing concern of antibiotic resistance.
