Phospho-EEF2K (Ser78) Antibody (PA5-143686) in WB
Western blot of Phospho-EEF2K (Ser78) in C2C12 cells treated with calyculin A (100 nM) for 30 minutes (lanes 1 & 3), then the blot was treated with lambda phosphatase (lanes 2 & 4). The blot was probed with anti-eEF2K (N-terminus) at a dilution of 1:500 (lanes 1 & 2) and Phospho-EEF2K (Ser78) polyclonal antibody (Product # PA5-143686) at a dilution of 1:1,000.

This antibody detects a 105 kDa* protein corresponding to the molecular mass of eEF2K on SDS-PAGE immunoblots of mouse C2C12 and Jurkat cells treated with Calyculin A, and this reactivity is removed after lambda phosphatase treatment.

EEF2K is a highly conserved Serine/Threoning kinase in the calmodulin signaling pathway whose activity is increased in cancers including breast cancer. EEF2K (eukaryotic elongation factor 2 kinase) is a threonine kinase that regulates protein synthesis by controlling the rate of peptide chain elongation. Up activation, by a variety of upstream kinases including AMPK or TRPM7, EEF2K phosphorylates the elongation factor EEF2 at a single site. Then, EEF2K renders EEF2 unable to bind ribosomes and inactive - in turn, the rate of protein synthesis is reduced.
