ERK1 and ERK3 are members of the extracellular signal regulated kinase family within the mitogen activated protein kinase network. ERK1 is encoded by MAPK3 and functions as a classical kinase activated by the Ras Raf MEK cascade, controlling transcription factor activity, cell cycle progression, proliferation, differentiation, and survival through cytoplasmic and nuclear substrates. ERK3 is encoded by MAPK6 and is an atypical kinase with a distinct activation mechanism that relies on phosphorylation and protein stability, often forming complexes with MK5 also known as PRAK to regulate cell motility and stress responses. ERK1 exhibits robust oscillatory activation and nuclear shuttling upon growth factor stimulation, whereas ERK3 shows constitutive or context dependent regulation and subcellular localization patterns that differ from ERK1 and ERK2. Both kinases are implicated in cancer biology, developmental processes, and inflammatory signaling, and are evaluated with gene expression assays, phospho specific immunodetection, and functional studies using small molecule inhibitors or genetic perturbations.
