FLNA and FLNB encode filamin A and filamin B, large actin binding scaffold proteins that crosslink F actin into orthogonal networks and anchor signaling receptors and ion channels to the cytoskeleton. Each filamin forms a dimer through a C terminal domain and contains an N terminal actin binding module followed by multiple immunoglobulin like repeats that provide mechanical flexibility and sites for interaction with membrane proteins, small GTPases, kinases, and transcriptional regulators. FLNA is broadly expressed and crucial for cell migration, mechanosensing, platelet function, and neuronal development, whereas FLNB has prominent roles in skeletal patterning, chondrocyte differentiation, and tissue morphogenesis. Pathogenic variants in FLNA cause periventricular nodular heterotopia, cardiovascular anomalies, and platelet disorders, while FLNB mutations lead to skeletal dysplasias such as atelosteogenesis and boomerang dysplasia, highlighting nonredundant functions despite overlapping activities. Filamins serve as mechanotransducers that couple extracellular matrix forces to intracellular signaling and gene expression, and they are studied using actin dynamics assays, interaction proteomics, imaging of cytoskeletal architecture, and genetic perturbation in cellular and animal models.
