Piperacillin is a broad-spectrum beta-lactam antibiotic that is often combined with tazobactam to enhance its efficacy against beta-lactamase-producing bacteria. Developed by Lederle Laboratories, piperacillin is primarily used to treat serious infections caused by Gram-negative bacteria, including Pseudomonas aeruginosa. The gene encoding piperacillin is located on the bacterial chromosome, and its structure consists of a penicillin core with an extended side chain that enhances its spectrum of activity. Functionally, piperacillin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins, leading to cell lysis and death. It is particularly effective in treating intra-abdominal infections and febrile neutropenia when combined with amikacin, due to its broad coverage of Gram-positive and Gram-negative organisms.
