20 µg of soluble protein from Chlamydomonas reinhardii, were separated on 10% SDS-PAGE and blotted for 1 h to nitrocellulose. Plastid acyl-ACP desaturase encodes a soluble, nuclear-encoded enzyme that is imported into plastids via an N-terminal chloroplast transit peptide and functions predominantly in the chloroplast stroma, where de novo fatty acid synthesis occurs. The mature protein is a homodimeric, ferritin-like soluble desaturase composed largely of alpha-helices; each monomer harbors a buried mu-oxo-bridged diiron active site coordinated by conserved acidic and histidine residues characteristic of the acyl-ACP desaturase domain, supporting oxygen-dependent redox chemistry. Biologically, plastid acyl-ACP desaturase catalyzes the first committed desaturation step in plastids by introducing a cis double bond into saturated acyl chains attached to acyl carrier protein (classically converting stearoyl-ACP to oleoyl-ACP), using O2 and reducing equivalents delivered through the ferredoxin system; this reaction helps determine cellular pools of monounsaturated fatty acids that feed downstream membrane lipid assembly and polyunsaturated fatty acid production, and perturbation of enzyme activity alters oleic acid availability in plastids with documented consequences for lipid homeostasis and defense-signaling crosstalk.
