PSB27-1 encodes a small, nuclear-encoded photosystem II (PSII) family protein that is targeted to chloroplasts and accumulates on the luminal side of the thylakoid membrane, where it associates with PSII assembly/repair intermediates rather than the fully mature complex. In Arabidopsis, PSB27-1 is an intronless gene producing a chloroplast-localized protein whose solved higher-plant structure shows a compact, predominantly helical bundle consistent with a soluble luminal assembly factor rather than an integral membrane subunit. Functionally, PSB27-1 contributes to PSII maintenance by promoting efficient recovery from photodamage (photoinhibition), supporting the PSII repair cycle; mechanistic work in oxygenic phototrophs places Psb27 proteins at the luminal face of PSII in contact with the CP43 antenna subunit, consistent with a role in regulating late-stage assembly/repair events around the water-oxidizing apparatus and/or reaction-center turnover.
