psbA encodes the D1 reaction center protein of photosystem II (PSII), a core thylakoid membrane subunit essential for oxygenic photosynthesis in cyanobacteria, algae, and plants. D1 binds key redox cofactors, including P680 chlorophylls, pheophytin, the primary quinone acceptor QA, and the non-heme iron, and forms the charge separating heterodimer with D2, enabling water oxidation via the oxygen-evolving complex. Because D1 experiences high photochemical turnover and is a major target of photodamage, psbA is among the most highly expressed plastid genes, supporting rapid D1 replacement through the PSII repair cycle. Multiple psbA isoforms or alleles can be differentially expressed under light intensity, UV stress, temperature, or nutrient status, optimizing PSII stability and photoprotection. Regulation occurs at transcriptional and translational levels, with psbA mRNA abundance, ribosome loading, and cofactor availability tightly coordinated. Mutations in psbA affect herbicide (e.g., atrazine, diuron) binding, electron transfer efficiency, and resilience to light stress, directly impacting photosynthetic performance and organismal fitness.
