psbE is a plastid-encoded gene in oxygenic photosynthetic organisms (including chloroplasts of plants and cyanobacteria) that produces a very small integral thylakoid-membrane protein forming the alpha subunit of cytochrome b559 within photosystem II (PSII). The PsbE polypeptide is typically ~80-90 amino acids long and is dominated by a single membrane-spanning alpha helix; together with the PSII cytochrome b559 beta subunit, it binds a b-type heme cofactor via a conserved histidine axial ligand contributed by each subunit, creating a tightly associated cytochrome embedded in the PSII reaction-center core. Functionally, cytochrome b559 is required for normal PSII activity and stability: genetic disruption of psbE (often affecting the psbE/psbF unit) or mutation of the heme-ligating histidine abolishes PSII electron transfer, supporting an essential role in PSII assembly and/or maintenance, with additional evidence that cytochrome b559 participates in protective redox pathways that help limit photodamage during stress conditions.
