PSBO1 encodes a soluble, nuclear-encoded extrinsic photosystem II (PSII) lumenal protein that is targeted to chloroplasts and accumulates on the lumen-facing side of thylakoid membranes as a core component of the oxygen-evolving complex. The gene produces a precursor polypeptide that is processed after import, yielding a mature protein of ~33 kDa that belongs to the conserved PsbO/manganese-stabilizing protein family and lacks transmembrane helices, consistent with peripheral association to PSII rather than membrane integration. Functionally, PsbO1 is the major PsbO isoform in Arabidopsis and is required for efficient water-splitting: it stabilizes the catalytic Mn4CaO5 cluster and supports proper oxygen evolution, with loss-of-function mutants showing impaired PSII performance and defects in oxygen-evolving complex stability and Ca utilization.
