PSBO2 encodes a nuclear-encoded, chloroplast-targeted extrinsic subunit of photosystem II that accumulates on the luminal side of the thylakoid membrane as part of the oxygen-evolving complex. The gene produces an mRNA that is translated as a precursor polypeptide with an N-terminal chloroplast transit peptide that is removed upon import, yielding a soluble, predominantly beta-strand-rich lumenal protein of the conserved PsbO/manganese-stabilizing protein family that associates non-covalently with the PSII core rather than spanning the membrane. Functionally, PsbO2 contributes to efficient oxygen evolution by stabilizing the water-oxidizing Mn4CaO5 catalytic cluster and helping maintain the proper ionic and structural environment required for water splitting; in Arabidopsis, genetic analyses indicate that PsbO2 is the minor isoform relative to PsbO1 yet is important for PSII performance under light stress and has a specific role in regulating phosphorylation state and turnover/repair of the PSII reaction-center D1 protein, consistent with a contribution to PSII maintenance in addition to catalysis support.
