The RASGRP4 (RAS Guanyl Releasing Protein 4) gene encodes a guanine nucleotide exchange factor (GEF) that specifically activates RAS proteins by facilitating the exchange of GDP for GTP. Located on chromosome 19, RASGRP4 is expressed primarily in hematopoietic cells, including mast cells and certain myeloid cells. The protein contains a diacylglycerol (DAG)-binding C1 domain, a Ras exchange motif, and a CDC25 domain, which are essential for its activation and function. RASGRP4 plays a pivotal role in signal transduction pathways that regulate cell proliferation, differentiation, and survival. Dysregulation of RASGRP4 has been linked to various diseases, including inflammatory disorders and cancers, particularly those involving the hematopoietic system. Studies have shown that overexpression or mutations in RASGRP4 can lead to aberrant RAS signaling, contributing to oncogenesis and disease progression.
