RBOHD encodes a plant NADPH oxidase (respiratory burst oxidase homolog D) that localizes predominantly to the plasma membrane, where it catalyzes extracellular/apoplastic reactive oxygen species (ROS) production as part of rapid stress and immune signaling. The protein is a large multi-pass membrane flavocytochrome with a long cytosolic N-terminus containing two EF-hand Ca2+-binding motifs and regulatory regions targeted by phosphorylation, coupled to a C-terminal dehydrogenase module bearing conserved FAD- and NADPH-binding sites; between these cytosolic regions, multiple (classically six) transmembrane helices form the membrane anchor and support heme-binding elements characteristic of NOX-family enzymes that transfer electrons from cytosolic NADPH across the membrane to molecular oxygen to generate superoxide that can dismutate to hydrogen peroxide. Biologically, RBOHD is a major source of stimulus-induced ROS during diverse responses, including pattern-triggered immunity and abiotic stress, contributing to localized defense outputs (e.g., oxidative burst-associated cell wall and signaling changes) and enabling rapid cell-to-cell and systemic signal propagation in response to wounding, temperature shifts, high light, salinity, and related cues.
