RDR6 encodes a cytoplasmic RNA-dependent RNA polymerase that functions in plant RNA silencing, acting downstream of small-RNA targeting to convert single-stranded RNA templates into double-stranded RNA that is subsequently processed into secondary siRNAs to reinforce and spread post-transcriptional gene silencing, including trans-acting siRNA biogenesis and antiviral defense. The encoded protein is a eukaryotic-type RdRP with a conserved catalytic core characteristic of RNA polymerases, including the essential Asp residue required for polymerase activity, and it also exhibits terminal nucleotidyltransferase activity and primer-independent as well as primer-dependent initiation on RNA templates in vitro. Structurally, RDR6 is a large, multi-domain protein containing an N-terminal RNA recognition motif-like region and a C-terminal RdRP domain that supports dsRNA synthesis needed for amplification (transitivity) of silencing signals and robust maintenance of silencing across tissues.
