RPL33 encodes a small, basic cytosolic ribosomal protein that localizes predominantly to the nucleolus and cytoplasm as a structural constituent of the large (60S) ribosomal subunit, where it associates with rRNA within the mature 80S ribosome and in pre-ribosomal particles during assembly. The encoded protein is part of the eukaryotic L33e family and is typically a short polypeptide enriched in positively charged residues consistent with rRNA binding, with multiple transcript variants reported that generally preserve the same coding sequence. Functionally, RPL33 contributes to ribosome biogenesis and efficient joining of the 40S and 60S subunits during translation initiation, thereby supporting global protein synthesis; perturbation of L33-family proteins has been shown to compromise large-subunit maturation and subunit joining, with downstream effects on translational control.
