RPN6 is a regulatory particle subunit of the 26S proteasome in plants that helps assemble and stabilize the 19S cap, enabling recognition, unfolding, and translocation of ubiquitinated proteins into the 20S core for degradation. It contributes to proteostasis and regulated protein turnover across development, hormone signaling, and stress responses, influencing pathways such as immunity, growth, and senescence. RPN6 interacts with chaperones and other lid and base subunits to promote proper proteasome biogenesis and activity, and its expression adjusts with environmental cues like heat, drought, and oxidative stress. Perturbation of RPN6 impairs proteasome function, leading to accumulation of damaged or regulatory proteins and defects in growth, fertility, and stress tolerance.
