Ramy3D is a rice (Oryza sativa) nuclear gene encoding an alpha-amylase that enters the secretory pathway and functions extracellularly/apoplastically to hydrolyze internal alpha-1,4-glucosidic bonds in starch and related alpha-glucans, thereby mobilizing carbohydrate reserves to support growth and metabolism when sugars are limiting. The encoded enzyme is a typical plant alpha-amylase polypeptide of ~436 amino acids, produced with an N-terminal signal peptide and a conserved catalytic architecture characteristic of glycoside hydrolase family 13, including the central (beta/alpha)8-barrel catalytic domain and a C-terminal beta-sandwich domain that contribute to substrate binding and enzymatic stability. At the genomic level, RAmy3D is a single member of a multi-gene alpha-amylase family and is physically linked to the related RAmy3E locus; its transcript is detected in multiple tissues (notably germinating seeds and callus) and is strongly regulated by carbon status, being repressed by sugars and induced upon sugar starvation through promoter elements that mediate metabolic control of transcription.
