Rnr1 is a nuclear-encoded, protein-coding gene whose product localizes predominantly to the cytoplasm and functions as the major large (alpha) subunit of the class I ribonucleotide reductase (RNR) complex, with regulated redistribution toward the nucleus under DNA replication stress. Rnr1 is a soluble enzyme subunit that forms an alpha2 catalytic core and associates with the small (beta) subunits to assemble the active alpha2beta2 RNR holoenzyme, in which the small subunit supplies the essential diferric-tyrosyl radical cofactor while Rnr1 provides the catalytic site and allosteric nucleotide-binding sites that control overall activity and substrate specificity. Biologically, Rnr1 catalyzes the rate-limiting reduction of ribonucleoside diphosphates to deoxyribonucleoside diphosphates, thereby sustaining balanced dNTP pools required for nuclear and mitochondrial DNA replication and for DNA repair; consistent with this central role, RNR1 expression is cell-cycle regulated and is also responsive to checkpoint signaling and metabolic cues (including iron availability) that tune dNTP production to replication demand and genotoxic stress.
