RNA polymerase beta1 is a large, essential DNA-directed RNA polymerase subunit found in the cytoplasm/nucleoid-associated transcription machinery of bacteria (exemplified by Escherichia coli rpoB), where it assembles with the other core subunits to form the catalytic enzyme responsible for transcription. The protein is a high-molecular-weight, multi-domain polypeptide (about 1.3k amino acids in E. coli) that contributes to the architecture of the active-center cleft and nucleic-acid binding channel, helping position the DNA template and nascent RNA during RNA chain initiation and elongation; its N-terminus is enriched in both acidic and basic residues, consistent with roles in macromolecular interactions. Functionally, the beta1 subunit is integral to the enzymatic activity of RNA polymerase, enabling accurate, processive synthesis of RNA from a DNA template using ribonucleoside triphosphates, thereby supporting global gene expression programs required for bacterial growth and stress responses.
