SAG12 (senescence-associated gene 12) encodes a plant papain-like cysteine endopeptidase that is developmentally induced at late stages of tissue aging, with transcript accumulation largely restricted to senescing organs and repressed when developmental senescence is delayed by factors such as cytokinins, auxin, or sugar status. The gene produces a secretory-pathway protease precursor with an N-terminal signal peptide followed by an autoinhibitory prodomain typical of C1A (papain family) proteases, yielding a mature catalytic domain that carries the conserved cysteine peptidase active-site features; the protein has been experimentally localized to senescence-associated vacuoles, specialized acidic lytic compartments that arise in senescing leaf cells and are distinct from the central vacuole. Functionally, SAG12 is thought to contribute to macromolecule breakdown during senescence-supporting nutrient (notably nitrogen) remobilization for reproductive growth-by promoting proteolysis within these senescence-associated vacuoles, and its strong senescence-specific induction has made it a widely used molecular marker and promoter for senescence-targeted transgene expression.
