SGS3, also called Suppressor of Gene Silencing 3, is a plant RNA binding protein essential for RNA directed DNA methylation and posttranscriptional gene silencing. It stabilizes double stranded RNA precursors by binding and protecting 3 prime overhangs, enabling RDR6 to synthesize secondary small interfering RNAs that amplify silencing signals. SGS3 localizes to cytoplasmic siRNA bodies and works with RDR6, DCL4, and AGO proteins to target viral RNAs, transposons, and aberrant endogenous transcripts. Its activity influences developmental regulation, antiviral defense, and stress responses by shaping small RNA populations and epigenetic marks. Loss of SGS3 impairs transgene and antiviral silencing, alters tasiRNA pathways, and leads to defects in leaf polarity, phase transitions, and resilience due to compromised small RNA biogenesis.
