Specific Species Reactivity: Arabidopsis thaliana
In Arabidopsis thaliana, SQE1, SQE2, and SQE3 encode closely related squalene epoxidases that are associated with endomembranes and catalyze the oxygenation of squalene to 2,3-oxidosqualene, a key gateway metabolite for cyclic triterpenoid production. SQE3 (a multi-exon gene) and SQE1 proteins localize to the endoplasmic reticulum, consistent with their role in channeling flux through sterol and brassinosteroid biosynthesis, while SQE2 is also enzymatically competent in heterologous assays but appears to contribute less to bulk squalene epoxidase activity in planta. Genetic evidence indicates these enzymes are only partially redundant: loss of SQE1 causes strong developmental phenotypes (including reduced root and hypocotyl elongation and defective seed development) and leads to squalene accumulation, demonstrating that SQE1 activity is essential for normal growth; SQE3 can functionally complement SQE1 and is particularly important for embryo development, whereas SQE2 does not compensate to the same extent. Collectively, SQE1-3 form a core enzymatic module in the mevalonate-derived isoprenoid pathway that supports membrane sterol homeostasis and downstream signaling/metabolic outputs dependent on oxidosqualene-derived triterpenoids.
