SRP43 encodes a chloroplast-localized, soluble signal recognition particle subunit that accumulates in the stroma and functions at the thylakoid membrane during post-translational targeting. The protein is a plant-specific multidomain factor of ~43 kDa built from a central ankyrin-repeat region and an N-terminal chromodomain module (three chromodomains have been described in canonical cpSRP43 proteins), creating multiple linear-motif binding surfaces for partner recognition. Functionally, SRP43 is a core component of the chloroplast SRP pathway that forms a soluble transit complex with cpSRP54 and directly binds light-harvesting chlorophyll a/b-binding proteins (LHCPs) to prevent their aggregation after import into the chloroplast; the ankyrin repeats mediate high-affinity recognition of an LHCP motif and are sufficient for the ATP-independent chaperone activity, while chromodomain-dependent interactions contribute to docking and coordination with other pathway components such as the Alb3 insertase and cpSRP54 to promote efficient delivery and insertion of LHCPs into the thylakoid membrane.
