Transmembrane protein 108 (TMEM108) is a gene associated with psychiatric disorders such as schizophrenia, bipolar disorder, and major depression disorder. It is specifically expressed in the nervous system, particularly enriched in the postsynaptic density fraction and DG granule neurons. Research has shown that TMEM108 is critical for adult neurogenesis in the hippocampal dentate gyrus, indicating its role in neuronal development and function. Studies with TMEM108 mutant mice exhibit mania-like behavior, highlighting its involvement in the pathology of bipolar disorder. The gene's expression is notably higher in oligodendrocyte lineage cells within the central nervous system compared to other lineage cells. As a schizophrenia susceptibility gene, TMEM108 has been identified as a potential target for understanding the underlying mechanisms of psychiatric diseases and for developing therapeutic approaches. However, the detailed pathophysiological mechanisms by which TMEM108 contributes to such disorders remain largely unknown.
