TOR genes encode large, conserved phosphatidylinositol 3-kinase-related serine/threonine protein kinases that localize predominantly to endomembrane surfaces, with TOR1 found on organelle-bounding membranes including the vacuolar membrane and also reported in the nucleus and at the plasma membrane, while TOR2 localizes to the vacuolar and plasma membranes; both act as catalytic subunits within multiprotein TOR complexes. Structurally, Tor proteins are approximately 280 kDa and are organized with extensive N-terminal HEAT repeat-rich regions that support scaffolding and complex assembly, followed by a central FAT region and an FKBP12-rapamycin-binding (FRB) module that mediates rapamycin sensitivity in TORC1, and a C-terminal protein kinase domain characteristic of PIKK family kinases capped by a short FATC tail important for activity and membrane association. Functionally, TOR integrates nutrient and growth-factor cues to coordinate cellular growth programs: TORC1 (containing Tor1 or Tor2) promotes anabolic metabolism by stimulating translation, transcriptional programs, and ribosome biogenesis while repressing catabolic responses such as autophagy, whereas TOR2 additionally supports TORC2-specific outputs that are rapamycin-insensitive and are required for plasma-membrane homeostasis and polarization of the actin cytoskeleton, thereby linking environmental status to cell-cycle progression and morphogenesis.
